SCIENTIFIC PRINCIPLES IN RELATIONSHIP DETERMINATION 

 

 INTRODUCTION

  • For relationship testing main equipment needed are PCR thermocyclers, Genetic analyzer like ABI capilary electrophoresis 3130 and the whole basic equipment complex .
  • CGAT has carried out thousands of experiments to improve and adapt each  step of  the protocols.
  • CGAT staff have determined allelic frequencies of 50 polymorphic loci in Vietnamese.
  • Our tests nowadays have accuracy more than 99,9999 %.

BASIC STEPS OF THE PROCESS

1. Sampling

Samples are gathered with a buccal swab, in which a swab is rubbed along the inside of the cheek to gather cells, or by drawing a small amount of blood.

 

 

2. DNA Extraction

   Human DNA is isolated by routine protocols from small amount of different samples, including fresh and dried blood, buccal swabs, tissue samples, semen etc....

 

 

3.  Specific DNA Multiplication 

  The DNA sequences of interest are multiplied in a thermocycler. As a result, the DNA sequences, representing different probes on different chromosomes are obtained in sufficient amounts to be studied.

 

 

4. Polyacrylamide Electrophoresis

    Electrophoresis is used for separation of DNA fragments with different size, which to be used for further considerations. At the moment we run both polyacrylamide and capillary electrophoresis.

 

 

5. Capilary Electrophoresis

   Most of DNA relationship experiments now are carried out by capillary electrophoresis with ABI sequencers...

 

 

6. Result Analysis 

    The final results look as the picture at the left. This is the allele report, on which the DNA fragments are indicated. The allele reports of different individuals are compared to each other to determine their genetic relationship.
